Kisspeptin is a naturally occurring peptide encoded by the KISS1 gene. It was first discovered in 1996 as a metastasis suppressor, but its critical role in reproductive biology became clear when researchers identified it as the master regulator of the hypothalamic-pituitary-gonadal (HPG) axis.
Kisspeptin binds to the GPR54 receptor (also called KISS1R) on GnRH neurons in the hypothalamus, triggering a cascade that stimulates the release of gonadotropin-releasing hormone (GnRH). This in turn signals the pituitary gland to release luteinizing hormone (LH) and follicle-stimulating hormone (FSH), which drive testosterone production in males and regulate ovulation and estrogen production in females.
